Zootec.
Guimarães,Simone Eliza Facioni; Euclydes,Ricardo Frederico.

Polymorphism in the internal transcribed spacer (ITS) of the ribosomal DNA of 26 isolates of ectomycorrhizal fungi Genet. Mol. Biol.
Gomes,Eliane A.; Kasuya,Maria Catarina M.; Barros,Everaldo G. de; Borges,Arnaldo C.; Araújo,Elza F..
Inter- and intraspecific variation among 26 isolates of ectomycorrhizal fungi belonging to 8 genera and 19 species were evaluated by analysis of the internal transcribed sequence (ITS) of the rDNA region using restriction fragment length polymorphism (RFLP). The ITS region was first amplified by polymerase chain reaction (PCR) with specific primers and then cleaved with different restriction enzymes. Amplification products, which ranged between 560 and 750 base pairs (bp), were obtained for all the isolates analyzed. The degree of polymorphism observed did not allow proper identification of most of the isolates. Cleavage of amplified fragments with the restriction enzymes Alu I, Hae III, Hinf I, and Hpa II revealed extensive polymorphism. All eight genera...

Genetic variation among pathogens causing "Helminthosporium" diseases of rice, maize and wheat Trop. Plant Pathol.
WEIKERT-OLIVEIRA,RITA C. B.; RESENDE,M. APARECIDA DE; VALÉRIO,HENRIQUE M.; CALIGIORNE,RACHEL B.; PAIVA,EDILSON.
Twenty isolates of four fungal species, agents of "Helminthosporium" diseases in cereals, were collected from different regions: nine Bipolarisoryzae isolated from rice (Oryza sativa), seven B.sorokiniana from wheat (Triticum aestivum), two B. maydis, and two Exserohilumturcicum from maize (Zea mays). The strains were compared by PCR-RFLP and RAPD analysis. Size polymorphism among the isolates in the ITS region comprising the 5.8 S rDNA indicated genetic differences among the isolates, while a UPGMA phenogram constructed after the digestion of this region with restriction enzymes showed inter- and intra-specific polymorphism. The RAPD profiles indicated an expressive level of polymorphism among different species, compared with a low level of polymorphism...

Genotypic diversity among brazilian isolates of Macrophomina phaseolina revealed by RAPD Trop. Plant Pathol.
Almeida,Álvaro M. R.; Abdelnoor,Ricardo V.; Arias,Carlos A. Arrabal; Carvalho,Valdemar P.; Jacoud Filho,David S.; Marin,Silvana R. R.; Benato,Luís C.; Pinto,Mauro C.; Carvalho,Cláudio G. P..
Macrophomina phaseolina has been considered one of the most prevalent soybean (Glycine max) pathogens in Brazil. No genetic resistance has been determined in soybean and very little is known about the genetic diversity of this pathogen in tropical and sub-tropical regions. Fifty-five isolates from soybean roots were collected in different regions and analyzed through RAPD for genetic diversity. The UPGMA cluster analysis for 74 loci scored permitted identification of three divergent groups with an average similarity of 99%, 92% and 88%, respectively. The three groups corresponded to 5.45%, 59.95% and 34.6%, respectively of all isolates used. A single plant had three different haplotypes, while 10.9% of the analyzed plants had two different haplotypes. In...

Detection of Metarhizium anisopliae var. anisopliae within infected sugarcane borer Diatraea saccharalis (Lepidoptera, Pyralidae) using specific primers Genet. Mol. Biol.
Destéfano,Ricardo Henri Rodrigues; Destéfano,Suzete A. Lanza; Messias,Claudio Luiz.
In order to construct specific primers for the detection and identification of the entomopathogenic fungus Metarhizium within infected sugarcane borer (Diatraea saccharalis) larvae we analyzed the ITS1 -5.8S- ITS2 rDNA regions of strains and varieties of M. anisopliae, M. album and M. flavoviride. The PCR amplification of these regions yielded a unique fragment of approximately 540 bp for M. anisopliae variety anisopliae strains E9, B/Vi and C (isolated in Brazil), 600 pb for M. a. anisopliae strain 14 (isolated in Australia), 650 bp for the M. album and 600 bp for M. flavoviride strains. The PCR products were digested with different restriction endonucleases (Afa I, Alu I, Dde I, Hae III, Hpa II and Sau 3A) and the PCR-RFLP profiles showed clear...

Molecular characterization of enteroinvasive Escherichia coli ipa genes by PCR-RFLP analysis BJM
Gibotti,Adriana; Tanaka,Tânia L.; Oliveira,Valéria R.; Taddei,Carla R.; Martinez,Marina B..
In this study, polymorphism in ipa genes was found in five out of nine EIEC serotypes studied. When SalI and HindII were used in RFLP-PCR assays many EIEC serotypes showed polymorphism in ipaB and ipaD. On the other hand, no polymorphism was observed in ipaA and ipaC in these strains. The polymorphism present in EIEC strains is serotype-dependent, since restriction patterns were conserved amongst strains belonging to the same serotype. When IpaB deduced amino acid sequences of S. flexneri M90T and FBC124-13 were compared, ten amino acids changes could be observed mainly in the amino-terminal region. The deduced EIEC IpaD amino-acid sequence presented 91% similarity with the Shigella strain. In this case, amino acid changes were spread out through the whole...

Kappa-casein gene study with molecular markers in female buffaloes (Bubalus bubalis) Genet. Mol. Biol.
Otaviano,Antonio Roberto; Tonhati,Humberto; Sena,Janete Aparecida Desidério; Cerón Muñoz,Mario Fernando.
Caseins comprise make up about 80% of the total protein content of milk and present polymorphism with changes in the amino acid sequence. Within this abundance of proteins, kappa-casein is noteworthy, since it has been associated with differences in milk yield, composition and processing. The objective of this study was to observe the existence of polymorphism in the kappa-casein gene in female buffaloes. For this purpose, blood samples from 115 female buffaloes, collected with vacutainer by needle punctionure of the jugular vein, were used. for genomic DNA extraction was done from blood samples. The PCR-RFLP and SSCP techniques demonstrated that the studied animals were monomorphic for the kappa-casein gene. Only allele B was observed in these animals,...

The use of PCR-RFLP as an identification tool for two closely related species of bats of genus Platyrrhinus Genet. Mol. Biol.
Ferreira,Juliana Machado; Martins,Felipe de Melo; Ditchfield,Albert; Morgante,João Stenghel.
The bat species Platyrrhinus lineatus and P. recifinus (Phyllostomidae: Stenodermatinae) are ecologically important because of their capacity for seed dispersal. P. recifinus is endemic to the Atlantic rain forest and is considered vulnerable by the IUCN. The lack of distinct morphological features makes identification of the two species a difficult task. This study was aimed at testing the hypothesis that these are actually two distinct species by using PCR-RFLP of the mitochondrial cytocrome b gene. The results showed no shared haplotypes, demonstrating that these are, in fact, two distinct species. No polymorphism was obtained for P. recifinus, which could be a sign of low genetic diversity in this threatened species.

DNA polymorphism of leptin gene in Bos indicus and Bos taurus cattle Genet. Mol. Biol.
Choudhary,Vivek; Kumar,Pushpendra; Bhattacharya,Tarun K.; Bhushan,Bharat; Sharma,Arjava.
Leptin plays an important role in the regulation of feed intake, energy metabolism, growth and reproduction of cattle. We used the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technique to screen for DNA polymorphisms of the leptin gene in 403 cattle belonging to various breeds of Bos indicus (Hariana, Sahiwal, Gir and Nimari cattle), Bos taurus (Holstein Friesian (HF) and Jersey cattle) as well as Bos taurus x Bos indicus crossbreds (½ HF x ½ Hariana). In all the cattle we amplified two regions of the leptin gene, a 522 bp fragment comprising the partial intron 2 and exon 3 and another 94 bp fragment consisting of part of exon 2. Digestion of 522 bp PCR products with the BsaAI restriction enzyme revealed three genotypes in...

Cryptosporidium infection in ostriches (Struthio camelus) in Brazil: clinical, morphological and molecular studies Rev. Bras. Ciênc. Avic.
Santos,MMAB; Peiró,JR; Meireles,MV.
Avian cryptosporidiosis has been reported in more than 30 species of birds. To date, the species infecting birds are C. baileyi, C. galli and C. meleagridis. In this study, the morphological, clinical and molecular characteristics of a Brazilian ostrich isolate of Cryptosporidium are described. The oocysts of this Brazilian isolate are larger and more elongated than those of Cryptosporidium previously reported in ostriches, which were morphologically similar to C. meleagridis. Morphological, biological and molecular analyses demonstrated similarity of this ostrich isolate with C. baileyi, suggesting that there are at least two Cryptosporidium species infecting ostriches; one with molecular, biological and morphological characteristics related to C....

Pathogenicity, molecular characterization, and cercosporin content of Brazilian isolates of Cercospora kikuchii Trop. Plant Pathol.
Almeida,Álvaro M. R.; Piuga,Fernanda F.; Marin,Silvana R. R.; Binneck,Eliseu; Sartori,Fábio; Costamilan,Leila M.; Teixeira,Maria R. O.; Lopes,Marcelo.
Cercospora kikuchii, involved with the defoliation of soybean (Glycine max) plants, is normally associated with Septoria glycines in late season. Seventy-two isolates from different regions in Brazil, obtained mainly from purple stained seeds, showed phenotypic variation. Cercosporin content and rate of colony growth was higly variable among isolates. A strong correlation between cercosporin content and virulence was identified. Genetic variation among and within populations was evaluated based on 86 RAPD loci. The RAPD analysis clustered all isolates into seven groups. No relationship was observed between RAPD groups and geographic origin or cercosporin content. The sequence of the internal spacer regions (ITS1-5.8S-ITS2) from 13 isolates chosen according...

A modified simple RFLP-PCR method for single nucleotide polymorphism (SNP) typing Genet. Mol. Biol.
Xiao,Junhua; Xin,Xiujuan; Luan,Xiaohui; Dongzhi,Wei; Shengli,Yang.
We describe a modified single nucleotide polymorphism (SNP) typing method based on the restriction fragment length polymorphism polymerase chain reaction (RFLP-PCR). This is a simple, economical method without the need for special equipment. For most SNP loci, a common restriction endonuclease (Hind III, EcoR I or BamH I) recognizing site (RER) can be introduced into one allelic form, but not the other by two rounds of mismatched PCR. The flanking regions can be changed by as many as five bases after PCR amplification with specially designed mismatching primers so the genotypes can be distinguished after digestion of the PCR products with corresponding endonucleases.
